Effects of culture conditions on the production of polyhydroxyalkanoates by Azotobacter chroococcum H23 in media containing a high concentration of alpechı́n (wastewater from olive oil mills) as primary carbon source

چکیده
Large amounts of homopolymers containing -hydroxybutyrate (PHB) and copolymers containing -hydroxyvalerate (P[HB-co-HV]) are produced by Azotobacter chroococcum strain H23 when growing in culture media amended with alpechı́n (wastewater from olive oil mills) as the sole carbon source. Copolymer was formed when valerate (pentanoate) was added as a precursor to the alpechı́n medium, but it was not formed with the addition of propionate as a precursor. A. chroococcum formed homoand copolymers of polyhydroxyalkanoates (PHAs) up to 80% of the cell dry weight, when grown on NH4 -medium supplemented with 60% (v/v) alpechı́n, after 48 h of incubation at 100 rev min and 30 °C. Production of PHAs by strain H23 using alpechı́n looks promising, as the use of a cheap substrate for the production of these materials is essential if bioplastics are to become competitive products. © 2002 Elsevier Science B.V. Production of Polyhydroxyalkanoates by Azotobacter chroococcum H23 in Wastewater from Olive Oil Mills (Alpechin)
We describe the production of large amounts of homoand copolymers of polyhydroxyalkanoates (PHAs) by Azotobacter chroococcum strain H23 when growing in culture media amended with alpechin. A. chroococcum grown on NH4+-medium supplemented with alpechin formed PHAs up to 50% of the cell dry weight after 24h. The results show that alpechin supports the growth of strain H23 and also that this waste...

Growth and utilization of different phenolic acids present in olive mill wastewater (OMW) by Azotobacter chroococcum were studied in chemically defined media. Growth and utilization of phenolic acids were only detected when the microorganism was cultured on p-hydroxybenzoic acid at concentration from 0.01% to 0.5% (w/v) and protocatechuic acid at concentration from 0.01% to 0.3% (w/v) as sole c...

Azotobacter vinelandii strain ATCC 12837 and A. chroococcum strain H23 (CECT 4435) were able to grow on N-free or NH4Cl-amended chemically-defined (Burk's) media, with protocatechuic acid (1-2 mmol 1(-1)) or sodium p-hydroxybenzoate (1-10 mmol 1(-1)) as sole carbon (C) sources. At a concentration of 2 mmol 1(-1), both substrates supported nitrogen fixation (acetylene reduction assay) at similar...
متن کاملInvestigating the amount of Soluble COD Removal from Wastewater Containing Olive Oil Using Electrocoagulation: A Study of Kinetic Models
Introduction and purpose: The olive oil industry wastewater contains significant amounts of oil and this can cause serious problems if it is discharged into the wastewater treatment system, so the oil in the wastewater must be removed and seperated prior to any action. Therefore, the oil in the sewage should be removed and separated prior to taking any measure. There are several ways to remove ...
متن کاملIsolation, Optimization, and Molecular Characterization of a Lipase Producing Bacterium from Oil Contaminated Soils
Lipases have many applications in biotechnology, thanks to their ability of acylglycerides hydrolysis. They alsp possess the unique feature of acting at the lipid-water interface, which distinguishes them from esterases. Commercially useful lipases are produced by microorganisms with the extracellular lipase being produced by many bacteria including Pseudomonas. The greatest production of lipas...
